Assessing risk behaviors and prevalence of sexually transmitted and blood-borne infections among female crack cocaine users in salvador - Bahia, Brazil BJID
Nunes,Ceuci L.X.; Andrade,Tarcisio; Galvão-Castro,Bernardo; Bastos,Francisco I.; Reingold,Arthur.
Crack cocaine use is associated with risky sexual behaviors and sexually transmitted infections (STIs), including HIV. We investigated sociodemographic and behavioral characteristics and infection rates in female crack cocaine users from impoverished communities of Salvador, Bahia, Brazil. A sample of 125 female crack cocaine users was recruited. Overall, the interviewees had low educational level and high rate of unemployment (close to 90%). One-third (37%) reported having traded sex for money or drugs, and 58% reported that they had not used condoms during intercourse in the last 30 days. The prevalence of infections was low: HIV-1.6%; HCV-2.4%; HBV- 0.8%; HTLV I/II-4.0%; and syphilis-4.0%. The combination of dire poverty and high prevalence of risk...

Distribution of the CCR2-64I allele in three Brazilian ethnic groups Genet. Mol. Biol.
Acosta,Angelina Xavier; Sampaio,Rogério Grimaldi; Spínola,Juliana Lima; Galvão-Castro,Bernardo.
CCR2 is a member of the superfamily of seven transmembrane domain G protein-coupled receptors, the largest receptor superfamily in the human genome. CCR2 acts as a receptor for MCP-1 (CC chemokine) and as a co-receptor for HIV-1 cell-target entry. The gene encoding this receptor is mapped to the chromosome band 3p21. A G-to-A transition at position 190 characterizes the CCR2-64I mutation, causing valine to isoleucine substitution in codon 64. This mutation has been identified as an important factor for delaying progression to AIDS. Here, we determined the prevalence of this allele in three different Brazilian populations: 261 Amerindians inhabiting an isolated region in northern Brazil (82 samples from the Waiampi tribe, and 179 samples from the Tiriyó...

Firmino,Alisson de Aquino; Martins,Adenilda Lima Lopes; Gois,Luana Leandro; Paixão,Taiane Silva; Batista,Everton da Silva; Galvão-Castro,Bernardo; Grassi,Maria Fernanda Rios.
ABSTRACT Introduction: Human T-cell lymphotropic virus type 1 (HTLV-1) is sexually transmitted and causes persistent infection. This virus induces activation of the immune system and production of inflammatory cytokines. This study aimed to assess the cytokine profile and cytopathological findings in the cervicovaginal fluid of asymptomatic HTLV-1-infected women. Methods: HTLV-1-infected and uninfected women were selected at the Centro de Atendimento ao Portador de HTLV in Salvador-Brazil. None of the included HTLV-1-infected women reported any HTLV-1-associated diseases. All volunteers underwent gynecological examination to collect cervicovaginal fluid. Cytokine quantification was performed using the Cytometric Bead Array (CBA) Human Th1/Th2/Th17 kit....

Carvalho,André Gordilho Joaquim de; Galvão-Phileto,Ana Verena; Lima,Nana Santos; Jesus,Rogério Santos de; Galvão-Castro,Bernardo; Lima,Manuela Garcia.
Viral infections and chronic diseases have been associated with psychiatric disorders. Among these, increased depression has been reported in HTLV-1 patients. However, no studies on the prevalence of other mental disturbances have been carried out in these patients. Salvador is the city with the highest rate of infection with HTLV-1 in Brazil and it is estimated that approximately 40,000 inhabitants are infected. In our cross sectional study, we examined the frequency of mental disturbances in 50 HTLV-1 seropositive patients followed at the Centro Integrativo e Multidisciplinar de HTLV e Hepatites Virais (CHTLV) of the Escola Bahiana de Medicina e Saude Pública (EBMSP) in Salvador from January to November 2007. We used a questionnaire to collect...

HLA-Bw4-B*57 and Cw*18 alleles are associated with plasma viral load modulation in HIV-1 infected individuals in Salvador, Brazil BJID
Silva,Edinete Melo da; Acosta,Angelina Xavier; Santos,Eduardo José Melo; Netto,Eduardo Martins; Lemaire,Denise Carneiro; Oliveira,Adriano Silva; Barbosa,Carolina Matos; Bendicho,Maria Teresita; Galvão-Castro,Bernardo; Brites,Carlos.
Host genetic factors play an important role in mediating resistance to HIV-1 infection and may modify the course of infection. HLA-B alleles (Bw4 epitope; B*27 and B*57) as well as killer cell immunoglobulin-like receptors have been associated with slow progression of HIV-1 infection. OBJECTIVE: To evaluate the association between serological epitopes HLA-Bw4 and HLA-Bw6 and prognostic markers in AIDS. METHODS: 147 HIV-infected individuals in Bahia, Northeast Brazil, were genotyped for HLA class I locus. HLA class I genotyping was performed by hybridization with sequence-specific oligonucleotide probes following amplification of the corresponding HLA-A, HLA-B and HLA-C genes. Statistical analysis was performed using Fisher's exact and ANOVA tests for...

Queiroz,Artur Trancoso Lopo de; Santos,Luciane Amorim; Moreau,Domingos Ramon; Oliveira,Tulio de; Watkins,David I.; Galvão-Castro,Bernardo; Alcantara,Luiz Carlos Junior.
Genetic analysis of HIV-1 is essential to improve treatment strategies and select epitopes for vaccine programs. The objective of this study was to determine whether known CD4+ and CD8+ epitopes were present in Brazilian HIV-1 strains. We used previously described CD8+ and CD4+ epitopes from the Los Alamos laboratory to search for these epitopes in the Brazilian sequences using the HIVbase program and we compared the frequency results with the analyses using physical-chemical profile tools from Network Protein Sequence Analysis (NPSA), and the SYFPEITHI program. Furthermore, this analysis was carried out with the Prosite tool using the GeneDoc program and ds/dn analyses using the Synonymous Nonsynonymous Analysis Program (SNAP). The HIVbase epitope mapping...

Study of the CCR5-m303 mutation in three different ethnic groups from Brazil Genet. Mol. Biol.
Grimaldi,Rogério; Acosta,Angelina Xavier; Cabral-Oliveira,Fernando Cézar; Brites,Carlos; Galvão-Castro,Bernardo.
The main coreceptor gene involved in HIV-1 infection is CCR5 beta chemokine receptor gene for which several mutations have been described, some of which have correlated with HIV-1 infection, acquired immune deficiency syndrome (AIDS), or both. Deletion of 32bp in the CCR5 gene (delta32) has been shown to confer resistance to infection by HIV-1 R5 strains. Another mutation, characterized by a thymine to adenine (T to A) nucleotide substitution at position 303 (m303), has shown the same effects as the delta32 mutation, with previous studies having shown that the allele frequency of the CCR5-m303 mutation is 0.014 in African-American and 0.007 in French populations. The Brazilian population is known to be genetically diverse, because of which we investigated...
